Bioinspired Molecular Recognition of DNA/RNA and Synthetic Polymers-Applications in Materials and Medicine

This talk will focus on a similar type of bioinspired molecular recognition in two very different environments. In the first part, the design and synthesis of small molecules that selectively recognize (CTG)-repeats in DNA and (CUG)-repeats in RNA will be described. CUG repeats are the causitive agent of an incurable disease called myotonic dystrophy type 1 (a form of muscular dystrophy). By targeting this sequence and developing cell permeable agents, we are working toward developing a therapeutic agent to treat this progressive disease.
In the second part of the talk, inspired by DNA we have synthesized and studied a artificial base-pairs that form hydrogen-bonded complexes with significantly higher stability and higher fidelity relative to the AT and GC base-pair.These novel base-pairs can act as nanoscale adhesive agents. Applications possible with these very high affinity recognition units will be highlighted. The agents have been particularly useful in enhancing the energetics at normally unfavorable interfaces at the molecular and materials level. They can serve as nanoscale glues in polymers and composites. Finally, the ability to turn the recognition on and off will be discussed.
